Pack labradoodle: prcd-PRA, rcd4-PRA, EIC, SD2, NEWS, vWD1, DM esone 2, CNM e HNPK

Pannello genetico multi-malattia per il Labradoodle (incrocio Labrador × Barboncino) che raggruppa nove test molecolari: due forme di atrofia progressiva della retina (prcd-PRA e rcd4-PRA), collasso indotto dall'esercizio (EIC), displasia scheletrica 2/nanismo (SD2), encefalopatia neonatale con convulsioni (NEWS), malattia di von Willebrand di tipo 1 (vWD1), mielopatia degenerativa (DM esone 2), miopatia centronucleare (CNM) e paraqueratosi nasale ereditaria (HNPK). Il pannello riunisce varianti descritte sia nel Labrador retriever sia nel Barboncino, il che riflette l'origine ibrida della razza. Il test molecolare è complementare all'esame oculare, neuromuscolare ed emostatico nella selezione riproduttiva.
Incidenza
Razza applicabile: Labradoodle (incrocio Labrador retriever × Barboncino). Le varianti provengono dalle razze parentali e non esistono stime pubblicate di frequenza dei portatori nella popolazione riproduttiva del Labradoodle (dati limitati). La prcd-PRA e la rcd4-PRA sono documentate nel Barboncino; l'EIC, la CNM, la SD2 e l'HNPK sono documentate nel Labrador; la DM esone 2 è ampiamente diffusa. La vWD1 da VWF c.7437G>A è documentata nel Barboncino nano e in altre razze (OMIA:001057-9615), ma non esiste uno studio specifico di razza nel Labradoodle; la sua reale rilevanza nella razza è incerta. In coorti ampie, la prcd-PRA rimane una delle varianti recessive più frequenti nelle razze di ascendenza retriever e barboncino (Donner et al., 2023; Clark et al., 2023).
Segni clinici
- Ridotta visione notturna e atrofia retinica progressiva (prcd-PRA e rcd4-PRA)\n- Cecità evolutiva con fondo oculare alterato\n- Debolezza dopo esercizio intenso con andatura barcollante e collasso (EIC)\n- Rapido recupero alla cessazione dell'esercizio (EIC)\n- Arti corti e conformazione nanica (SD2)\n- Convulsioni e deterioramento neurologico nel cucciolo neonatale (NEWS)\n- Morte precoce nella maggior parte dei casi (NEWS)\n- Sanguinamento prolungato dopo chirurgia o trauma, ecchimosi ed epistassi (vWD1)\n- Paresi progressiva degli arti posteriori con atassia propriocettiva (DM)\n- Debolezza muscolare e atrofia fin da giovani con andatura barcollante (CNM)\n- Ipercheratosi nasale con fissurazioni e crosta nasale (HNPK)
Storia
I test del pannello sono stati sviluppati in modo indipendente man mano che i gruppi di genetica canina caratterizzavano varianti specifiche per ciascuna malattia. La prcd-PRA è stata associata al gene PRCD (Zangerl et al., 2006). La rcd4-PRA è stata associata a una mutazione con slittamento del frame in C2orf71/PCARE, descritta nel Gordon setter e nell'Irish setter (Downs et al., 2013) e successivamente documentata nel Barboncino standard e nano e in altre razze (Downs et al., 2014; OMIA:001575-9615). Il collasso indotto dall'esercizio è stato associato a DNM1 nel Labrador retriever (Patterson et al., 2008). La displasia scheletrica 2 (SD2) è stata associata a COL11A2 nel Labrador (Frischknecht et al., 2013). L'encefalopatia neonatale con convulsioni del Barboncino standard (NEWS) è stata associata ad ATF2 (Chen et al., 2008). La vWD di tipo 1 è associata alla variante VWF c.7437G>A, descritta nel Dobermann e in più di una dozzina di razze (Crespi et al., 2018; OMIA:001057-9615). La miopatia centronucleare (CNM) del Labrador è stata associata a un'inserzione SINE in HACD1 (precedentemente PTPLA) (Pelé et al., 2005; Maurer et al., 2012). La paraqueratosi nasale ereditaria (HNPK) del Labrador è stata associata a SUV39H2 (Jagannathan et al., 2013). La DM canina è stata collegata alla variante dell'esone 2 di SOD1 (Awano et al., 2009).
Gestione dell'allevatore
- Genotipizzare i riproduttori prima dell'accoppiamento; il pannello copre nove condizioni con un solo campione\n- Per le condizioni recessive (prcd-PRA, rcd4-PRA, EIC, SD2, NEWS, CNM, HNPK, DM): non accoppiare due portatori — rischio del 25 % di omozigoti; portatore × indenne è sicuro per la discendenza destinata all'allevamento se testata\n- Per la DM: ricordare la penetranza incompleta; gli omozigoti non sono una priorità riproduttiva e una decisione clinica non deve basarsi solo sul genotipo\n- Per la vWD1 (VWF c.7437G>A; espressione dominante con penetranza incompleta): valutare il livello del fattore von Willebrand e confermare con il laboratorio la variante esatta impiegata nel pannello\n- Dopo un caso clinico confermato, non ripetere l'accoppiamento parentale e comunicare lo stato all'acquirente
Note dello specialista
Le due PRA del pannello si differenziano per età di esordio e fondo oculare, ma è la diagnosi molecolare che conferma il sospetto; la rcd4-PRA è dovuta a C2orf71/PCARE e non a CNGB3. Esame oculare annuale (ECVO/CERF) complementare. L'EIC deve essere differenziato dal collasso da cardiomiopatia aritmogena, miopatia laringea e crisi adinamica. La NEWS deve essere differenziata da altre encefalopatie neonatali (infettive, metaboliche, tossiche). La vWD1 si conferma con il dosaggio dell'antigene e dell'attività del fattore von Willebrand; la variante del pannello (VWF c.7437G>A) è documentata nel Barboncino nano, ma non specificamente nel Labradoodle. La CNM si conferma con biopsia muscolare (fibre centronucleari) e test molecolare. L'HNPK si conferma con esame clinico e test molecolare. La DM è diagnosi di esclusione: escludere la compressione midollare prima di attribuire il quadro a SOD1.
