Pack labradoodle: prcd-PRA, rcd4-PRA, EIC, SD2, NEWS, vWD1, DM exão 2, CNM e HNPK

Painel genético multidoença para o Labradoodle (cruzamento Labrador × Caniche) que agrupa nove testes moleculares: duas formas de atrofia progressiva da retina (prcd-PRA e rcd4-PRA), colapso induzido pelo exercício (EIC), displasia esquelética 2/nanismo (SD2), encefalopatia neonatal com convulsões (NEWS), doença de von Willebrand tipo 1 (vWD1), mielopatia degenerativa (DM exão 2), miopatia centronuclear (CNM) e paraqueratose nasal hereditária (HNPK). O painel reúne variantes descritas tanto no Labrador retriever como no Caniche, o que reflete a origem híbrida da raça. O teste molecular é complementar do exame ocular, neuromuscular e hemostático na seleção reprodutiva.
Incidência
Raça aplicável: Labradoodle (cruzamento Labrador retriever × Caniche). As variantes provêm das raças parentais e não há estimativas publicadas de frequência de portadores na população reprodutora do Labradoodle (dados limitados). A prcd-PRA e a rcd4-PRA estão documentadas no Caniche; a EIC, a CNM, a SD2 e a HNPK estão documentadas no Labrador; a DM exão 2 está amplamente difundida. A vWD1 por VWF c.7437G>A está documentada no Caniche miniatura e noutras raças (OMIA:001057-9615), mas não há estudo específico de raça no Labradoodle; a sua relevância real na raça é incerta. Em coortes amplas, a prcd-PRA continua a ser uma das variantes recessivas mais frequentes em raças de ascendência retriever e caniche (Donner et al., 2023; Clark et al., 2023).
Manejo do criador
- Genotipar os reprodutores antes da cobrição; o painel cobre nove condições numa só amostra\n- Para as condições recessivas (prcd-PRA, rcd4-PRA, EIC, SD2, NEWS, CNM, HNPK, DM): não cruzar dois portadores — risco de 25 % de homozigóticos; portador × livre é seguro para a descendência destinada à criação se for testada\n- Para a DM: recordar a penetrância incompleta; os homozigóticos não são prioridade reprodutiva e uma decisão clínica não deve basear-se apenas no genótipo\n- Para a vWD1 (VWF c.7437G>A; expressão dominante com penetrância incompleta): avaliar o nível do fator von Willebrand e confirmar com o laboratório a variante exata empregue no painel\n- Após um caso clínico confirmado, não repetir o cruzamento parental e comunicar o estatuto ao comprador
Notas do especialista
As duas PRAs do painel diferenciam-se pela idade de início e pelo fundo ocular, mas é o diagnóstico molecular que confirma a suspeita; a rcd4-PRA deve-se a C2orf71/PCARE e não a CNGB3. Exame ocular anual (ECVO/CERF) complementar. A EIC deve ser diferenciada de colapso por cardiomiopatia arritmogénica, miopatia laríngea e crise adinâmica. A NEWS deve ser diferenciada de outras encefalopatias neonatais (infecciosas, metabólicas, tóxicas). A vWD1 confirma-se por doseamento do antigénio e da atividade do fator von Willebrand; a variante do painel (VWF c.7437G>A) está documentada no Caniche miniatura, mas não especificamente no Labradoodle. A CNM confirma-se por biópsia muscular (fibras centronucleares) e teste molecular. A HNPK confirma-se por exame clínico e teste molecular. A DM é diagnóstico de exclusão: descartar compressão medular antes de atribuir o quadro a SOD1.
