Pack American bully: atrofia progresiva de retina (crd1-PRA) + cistinuria + hiperuricosuria (HUU/SLC) + lipofuscinosis neuronal ceroide (NCL) + mielopatía degenerativa (DM exón 2)

Panel genético del American bully que agrupa cinco condiciones hereditarias recesivas prevalentes en razas tipo bull/pit: la atrofia progresiva de retina tipo crd1 (crd1-PRA, degeneración precoz de fotorreceptores), la cistinuria (cálculos de cistina en vías urinarias), la hiperuricosuria/hiperuricemia (HUU, riesgo de urolitos de urato), la lipofuscinosis neuronal ceroide (NCL, degeneración neurodegenerativa de inicio adulto) y la mielopatía degenerativa (DM, variante del exón 2 de SOD1). El American bully desciende del American Staffordshire terrier y comparte varias variantes por efecto fundador.
Incidencia
American bully (y razas tipo pit/bull emparentadas). La NCL por ARSG está caracterizada en AmStaff/APBT (alelo al 3,4 % en AmStaff, Donner 2023 n>1M); se asume compartida con el American bully por ancestria, sin casos publicados en la raza. Para crd1-PRA, HUU y cistinuria en American bully las cifras poblacionales no se publican de forma fiable (datos limitados).
Manejo del criador
- Genotipar reproductores para PDE6B, SLC2A9, ARSG, SOD1 exón 2 y la variante de cistinuria antes de la cubrición
- No cruzar dos portadores para la misma variante
- Un portador puede cruzarse con un libre y la descendencia destinada a cría debe testarse
- Tras un caso confirmado, no repetir el cruce parental y comunicar el estatus al comprador
- Para HUU y cistinuria, recomendar dieta y manejo (baja purina, baja metionina) aunque el test sea positivo, y vigilancia urinaria
- Recordar que la DM tiene penetrancia incompleta: no todos los SOD1 homocigotos desarrollan enfermedad
- No cruzar dos portadores para la misma variante
- Un portador puede cruzarse con un libre y la descendencia destinada a cría debe testarse
- Tras un caso confirmado, no repetir el cruce parental y comunicar el estatus al comprador
- Para HUU y cistinuria, recomendar dieta y manejo (baja purina, baja metionina) aunque el test sea positivo, y vigilancia urinaria
- Recordar que la DM tiene penetrancia incompleta: no todos los SOD1 homocigotos desarrollan enfermedad
Notas del especialista
La crd1-PRA debe distinguirse de otras PRA (prcd-PRA, etc.); la prueba de crd1 no excluye otras formas. La NCL por ARSG tiene presentación adulta y debe diferenciarse de otras ataxias degenerativas. La cistinuria y la HUU comparten presentación clínica (urolitiasis) y requieren análisis del cálculo. La DM solo es atribuible a SOD1 tras descartar compresión medular (RM, mielografía).
