Pack Schnauzer Gigante: NECAP1-PRA, prcd-PRA, DCM, HUU/SLC e DM exão 2

Painel multidoença do Schnauzer Gigante que agrupa cinco testes moleculares: duas formas de atrofia progressiva da retina (NECAP1-PRA e prcd-PRA), cardiomiopatia dilatada (DCM por deleção de 22 pb em RBM20), hiperuricosúria (HUU, SLC2A9) e mielopatia degenerativa (DM, variante do exão 2 de SOD1). Combina afeções oculares, cardíacas, urinárias e neurológicas. O teste molecular é complementar, e não substitutivo, do exame ocular anual e do acompanhamento cardiológico na seleção reprodutiva.
Incidência
Raça aplicável: Schnauzer Gigante. DCM por RBM20: no Schnauzer standard ~21 % transportavam pelo menos um alelo (19,7 % heterozigotos; 1,5 % homozigotos) e no Schnauzer Gigante 9,8 % (Leach 2022); a variante não foi detetada nas outras raças analisadas. A HUU está descrita no Schnauzer Gigante a frequências baixas (Karmi et al., 2010; frequência por raça: Karmi et al., 2010, J Vet Intern Med, PMID: 21054540). A prcd-PRA e a DM são testes pan-raça sem evidência específica de raça publicada no Schnauzer Gigante; a NECAP1-PRA tem casuística escassa. Frequências fiáveis de portadores na população reprodutora: dados limitados.
Manejo do criador
- Genotipar os reprodutores antes da cobrição; o painel cobre cinco condições numa única amostra\n- Condições recessivas (NECAP1-PRA, prcd-PRA, HUU, DM): não cruzar dois portadores (25 % de homozigotos afetados); portador x livre é admissível se a descendência destinada à criação for testada\n- DCM (RBM20): não usar homozigotos como reprodutores e ponderar ecocardiografia e Holter nos reprodutores, além do teste molecular\n- HUU: em homozigotos, dieta baixa em purinas, hidratação abundante e vigilância urinária\n- Após um caso clínico confirmado, não repetir o cruzamento parental e comunicar o estado ao comprador
Notas do especialista
Exame ocular anual (ECVO) e eletrorretinografia quando aplicável: várias PRA são tardias. A DCM requer diagnóstico por ecocardiografia e Holter; o teste molecular não substitui o acompanhamento cardiológico, e a variante RBM20 não explica todos os casos de DCM da raça. A HUU é confirmada por urianálise e, se houver cálculos, por análise do cálculo (diferenciar de cistina e infeciosos). A DM é diagnóstico de exclusão (descartar compressão medular, hérnia discal e tumores).
