Pack Schnauzer gigante: NECAP1-PRA, prcd-PRA, DCM, HUU/SLC y DM exón 2

Panel multienfermedad del Schnauzer gigante que agrupa cinco pruebas moleculares: dos formas de atrofia progresiva de retina (NECAP1-PRA y prcd-PRA), cardiomiopatía dilatada (DCM por deleción de 22 pb en RBM20), hiperuricosuria (HUU, SLC2A9) y mielopatía degenerativa (DM, variante del exón 2 de SOD1). Combina afecciones oculares, cardíacas, urinarias y neurológicas. La prueba molecular es complementaria, no sustitutiva, del examen ocular anual y del seguimiento cardiológico en la selección reproductiva.
Incidencia
Raza aplicable: Schnauzer gigante. DCM por RBM20: en Schnauzer estándar ~21 % portaban al menos un alelo (19,7 % heterocigotos; 1,5 % homocigotos) y en Schnauzer gigante 9,8 % (Leach 2022); la variante no se detectó en otras razas analizadas. La HUU está descrita en el Schnauzer gigante a frecuencias bajas (Karmi et al., 2010; frecuencia por razas: Karmi et al., 2010, J Vet Intern Med, PMID: 21054540). La prcd-PRA y la DM son pruebas pan-raciales sin evidencia raza-específica publicada en el Schnauzer gigante; la NECAP1-PRA tiene casuística escasa. Frecuencias fiables de portadores en la población reproductora: datos limitados.
Manejo del criador
- Genotipar a los reproductores antes de la cubrición; el panel cubre cinco condiciones en una sola muestra
- Condiciones recesivas (NECAP1-PRA, prcd-PRA, HUU, DM): no cruzar dos portadores (25 % de homocigotos afectos); portador x libre es admisible si se testa la descendencia destinada a cría
- DCM (RBM20): no usar homocigotos como reproductores y valorar ecocardiografía y Holter en reproductores, además del test molecular
- HUU: en homocigotos, dieta baja en purinas, hidratación abundante y vigilancia urinaria
- Tras un caso clínico confirmado, no repetir el cruce parental y comunicar el estatus al comprador
- Condiciones recesivas (NECAP1-PRA, prcd-PRA, HUU, DM): no cruzar dos portadores (25 % de homocigotos afectos); portador x libre es admisible si se testa la descendencia destinada a cría

Notas del especialista
Examen ocular anual (ECVO) y electrorretinografía cuando proceda: varias PRA son tardías. La DCM requiere diagnóstico por ecocardiografía y Holter; el test molecular no sustituye el seguimiento cardiológico, y la variante RBM20 no explica todos los casos de DCM de la raza. La HUU se confirma por urianálisis y, si hay cálculos, por análisis del cálculo (diferenciar de cistina e infecciosos). La DM es diagnóstico de exclusión (descartar compresión medular, hernia discal y tumores).
