Pack Schnauzer gigante: NECAP1-PRA, prcd-PRA, DCM, HUU/SLC e DM esone 2

Pannello multi-malattia del Schnauzer gigante che raggruppa cinque test molecolari: due forme di atrofia progressiva della retina (NECAP1-PRA e prcd-PRA), cardiomiopatia dilatativa (DCM da delezione di 22 pb in RBM20), iperuricosuria (HUU, SLC2A9) e mielopatia degenerativa (DM, variante dell'esone 2 di SOD1). Combina patologie oculari, cardiache, urinarie e neurologiche. Il test molecolare è complementare, non sostitutivo, dell'esame oculare annuale e del monitoraggio cardiologico nella selezione riproduttiva.
Incidenza
Razza applicabile: Schnauzer gigante. DCM da RBM20: nello Schnauzer standard ~21 % portavano almeno un allele (19,7 % eterozigoti; 1,5 % omozigoti) e nel Schnauzer gigante 9,8 % (Leach 2022); la variante non è stata rilevata nelle altre razze analizzate. La HUU è descritta nel Schnauzer gigante a basse frequenze (Karmi et al., 2010; frequenza per razza: Karmi et al., 2010, J Vet Intern Med, PMID: 21054540). La prcd-PRA e la DM sono test pan-razza senza evidenza razza-specifica pubblicata nel Schnauzer gigante; la NECAP1-PRA ha casistica scarsa. Frequenze affidabili dei portatori nella popolazione riproduttiva: dati limitati.
Gestione dell'allevatore
- Genotipizzare i riproduttori prima dell'accoppiamento; il pannello copre cinque condizioni in un unico campione\n- Condizioni recessive (NECAP1-PRA, prcd-PRA, HUU, DM): non incrociare due portatori (25 % di omozigoti affetti); portatore x libero è ammissibile se la discendenza destinata all'allevamento viene testata\n- DCM (RBM20): non usare omozigoti come riproduttori e valutare ecocardiografia e Holter nei riproduttori, oltre al test molecolare\n- HUU: negli omozigoti, dieta a basso contenuto di purine, idratazione abbondante e sorveglianza urinaria\n- Dopo un caso clinico confermato, non ripetere l'incrocio parentale e comunicare lo stato all'acquirente
Note dello specialista
Esame oculare annuale (ECVO) ed elettroretinografia quando opportuno: diverse PRA sono tardive. La DCM richiede diagnosi mediante ecocardiografia e Holter; il test molecolare non sostituisce il monitoraggio cardiologico, e la variante RBM20 non spiega tutti i casi di DCM della razza. La HUU è confermata da urianalisi e, se ci sono calcoli, dall'analisi del calcolo (differenziare da cistina e infettivi). La DM è diagnosi di esclusione (escludere compressione midollare, ernia del disco e tumori).
