Pack Kromfohrländer: vWD1, Hiperqueratosis digital (DH/HFH), HUU/SLC, DM exón 2, MDR1 y Furnishing

Panel genético multienfermedad del Kromfohrländer que agrupa seis pruebas moleculares: enfermedad de von Willebrand tipo 1 (vWD1), hiperqueratosis digital hereditaria (HFH, FAM83G), hiperuricosuria (HUU, SLC2A9), mielopatía degenerativa (DM, variante del exón 2 de SOD1), variante del gen ABCB1/MDR1 (sensibilidad farmacológica) y furnishing (pelo duro/cejas, RSPO2). Combina afecciones clínicas con un marcador farmacogenético y un carácter de capa. El panel es complementario, no sustitutivo, del examen clínico y del seguimiento de la cría.
Incidencia
Raza aplicable: Kromfohrländer. La variante FAM83G c.155G>C está descrita en Kromfohrländer e Irish terrier (y Bedlington terrier); la serie poblacional del Dogo de Burdeos (KRT16, base molecular distinta) mostró ~20 % de portadores, pero no es extrapolable al Kromfohrländer. La HUU, la DM, el defecto MDR1 y el furnishing son pruebas pan-raciales sin evidencia publicada específica en el Kromfohrländer (datos limitados). La vWD1 y la HFH (FAM83G) sí están documentadas en la raza. No hay frecuencias fiables de portadores en la población reproductora del Kromfohrländer.
Manejo del criador
- Genotipar a los reproductores con el panel antes de la cubrición, usando la prueba de la variante correspondiente a la raza
- vWD1: autosómica dominante con penetrancia incompleta (documentada en la raza, Segert 2019); valorar el cruce con un ejemplar libre y complementar con la determinación del nivel de vWF
- HFH, HUU y DM (recesivas): no cruzar dos portadores (25 % de homocigotos afectos); portador x libre es admisible si se testa la descendencia destinada a cría
- MDR1 (efecto dosis): no cruzar dos portadores; informar al comprador y al veterinario para evitar ivermectina, moxidectina, loperamida y otros sustratos de P-gp en homocigotos y heterocigotos
- Furnishing: usar la prueba como selección de capa, no como cribado de enfermedad, e interpretarla junto con otros loci de capa
- Tras un caso clínico confirmado, no repetir el cruce parental
- vWD1: autosómica dominante con penetrancia incompleta (documentada en la raza, Segert 2019); valorar el cruce con un ejemplar libre y complementar con la determinación del nivel de vWF

Notas del especialista
La vWD1 se confirma por dosaje de antígeno y actividad del factor de von Willebrand; valorar antes de cirugía programada. La HFH se confirma por clínica y test molecular (o biopsia); el diferencial incluye parakeratosis nasal del Labrador (SUV39H2), ictiosis epidermolítica del Norfolk terrier, leishmaniosis, pémfigo y déficit de zinc. La urolitiasis por urato debe diferenciarse de la cistina e infecciosa (análisis del cálculo). La DM es diagnóstico de exclusión (descartar compresión medular). No aplicar lenguaje médico de afecto/portador al furnishing.
